Fluorescent “superheroes”! mBaoJin + mScarlet3-H (also known as mYongHong) Tool Viruses Empower Super-Resolution and High-Efficiency Imaging

In cutting-edge research fields such as cell biology and neuroscience, the brightness, stability, and specificity of fluorescent labels directly determine the experimental limits. Brain Case Biotech, with years of deep expertise in the field of viral tools, is proud to announce the launch of our new viral vector products. These tools integrate two top-tier fluorescent proteins—mBaoJin Green Fluorescent Protein (developed by the Kiryl Piatkevich team at Westlake University's School of Life Sciences) and mScarlet3-H Red Fluorescent Protein (developed by the Zhifei Fu team at Fujian Medical University)—into our optimized viral delivery systems.By combining the advantages of "superior photostability + precise delivery," our viral vectors effectively overcome the limitations of traditional imaging techniques. This breakthrough provides researchers with a more powerful and reliable visualization tool, pushing the boundaries of discovery.

The Dynamic Duo of Fluorescent Proteins, Performance Crushing Traditional Probes

01 mBaoJin: The Green Champion with High Brightness and Anti-Photobleaching

As an optimized version of StayGold monomer (Nat Methods. 2024 Apr;21(4):657-665.,), mBaoJin perfectly inherits the high brightness advantage and achieves an all-around performance upgrade:
✅ Pure Monomer, No Interference: With an OSER experiment score of 93.7%, it seamlessly integrates with target proteins, avoiding positional deviations caused by dimerization, making it ideal for detailed studies such as organelle labeling and protein dynamics tracking.

✅ Fast Maturation and Acid-Base Resistance: In a physiological environment at 37°C, the maturation half-life is only 7.5 minutes, 2.2 times faster than mNeonGreen. It has a pKa as low as 4.37, maintaining stable fluorescence even in acidic cellular environments.

✅ Super Strong Photostability: Its photostability under low excitation intensity is 15 times that of eGFP and 9 times that of mNeonGreen, supporting live cell super-resolution imaging for up to 60 minutes, with significantly reduced phototoxicity.

✅ Exceptional Chemical Compatibility: It withstands harsh treatments like 6M guanidine hydrochloride and PFA fixation, perfectly compatible with expansion microscopy (ExM), enabling high-resolution imaging of subcellular structures.

02 mScarlet3-H (mYongHong): The All-Purpose Red Benchmark with Unmatched Durability

An upgraded masterpiece derived from mScarlet3 (Nat Methods. 2025 Jun;22(6):1288-1298, Nature Methods | mYongHong: The Next-Generation Ultra-Stable Monomer Red Fluorescent Protein), mYongHong redefines the standard for red fluorescent proteins with its extreme stability:
✅ Extreme Environment Tolerance: After being treated at 90°C for 1 hour, it retains 82.5% fluorescence, with a 500% increase in resistance to key CLEM reagent OsO₄, providing a signal-to-noise ratio 3 times higher than traditional probes.

✅ Anti-Photobleaching and Quenching: Its photostability meets the requirements for STED super-resolution imaging, with less than 5% fluorescence loss after 2 hours of continuous imaging, far surpassing traditional RFPs like mCherry and tdTomato.

✅ Rapid Clearing Adaptation: Supports 60°C accelerated tissue clearing, completing whole mouse brain clearing in 24 hours with 68% fluorescence retention, significantly shortening experimental timelines.

✅ Monomeric High Compatibility: Its pure monomeric structure prevents aggregation, making it highly compatible with viral vector long-term expression without interfering with normal cellular functions, ideal for neural circuit tracing and in vivo dynamic observations.

01 Flexible Choice of Multiple Vectors

Recombinant Adeno-Associated Virus (rAAV): Strong neurotropism, supports various promoters like hSyn, CMV, and is suitable for neural circuit tracing and long-term in vivo imaging.

Lentivirus (LV): Wide infection spectrum, integrates into the host genome for stable fluorescence marking, ideal for cell line labeling and protein expression tracking.

Retrograde Virus (RV/AAV-Retro):
 Precisely enables retrograde neural circuit tracing, with dual fluorescent labeling for clear analysis of synaptic connections.



02  Customized Research Solutions
Dual Fluorescent Labeling Combination: mBaoJin (green) + mScarlet3-H (red), with non-overlapping spectra, supports multi-target tracking simultaneously and is compatible with various microscopes, including wide-field, confocal, and super-resolution.

Specific Targeting: Combines tissue-specific promoters with viral serotype selection to achieve precise labeling of specific cell types, such as neurons, glial cells, and cardiomyocytes.
