A modified variant of the 5-hydroxytryptamine type 3 receptor (5-HT3R) that incorporates the pharmacological properties of Proline-Specific Adenosine Methyltransferase (PSAM) for chemogenetic control. It is composed of 5-HT3R subunits with PSAM4-specific mutations introduced, enabling specific binding to PSAM ligands and generating ion channel activity. PSAM4-5HT3 retains the cation-selective conductance properties of the native 5-HT3 receptor and depolarizes the cell membrane upon activation by the agonist varenicline, thereby allowing neuronal activation.
